The phrase is usually said by an armed man during a robbery or arrest.

The verb "stick" can be substituted for the verb "put". 

"Em" is a slang abbreviation for "them".

Used to order someone to raise his/hands above their shoulders.

"Put 'em up!" shouted the gunman.

When a robber said "Put 'em up", you should raise both hands in the air.
